𝐂𝐨𝐧𝐟𝐢𝐫𝐦𝐚𝐭𝐨𝐫𝐲 𝐓𝐞𝐬𝐭 𝐅𝐨𝐫 𝐁𝐥𝐨𝐨𝐝

Microcrystal Test
  • In microcrystal test, chemicals are applied to dried blood stains which target the non-protein heme group of hemoglobin, the oxygen carrying protein of RBCs that belongs to class of compound known as porphyrins.
  • These assays cannot distinguish between human and animal blood.
Teichmann’s test
  • Also known as Hematin Crystal Assay.
  • Teichmann’s reagent:
  1. KCl, KBr and KI 0.1 g each
  2. Glacial Acetic Acid 100ml
Procedure:
  • Suspected blood sample is placed on a microscopic slide.
  • Then glacial acetic acid is added to the slide with small amount of chloride which is followed by heating.
  • As a result, crystals of ferriprotoporphyrin chloride are formed.
  • Crystals are rhombic in shape and are brown in colour.

Takayama Test
  • It is also known as Haemochromogen crystal assay test.
  • Takayama's reagent: 
  1. Saturated solution of glucose 3 ml
  2. Pyridine solution 3 ml
  3. 10% Sodium Hydroxide Solution 3 ml
  4. Glacial acetic acid 7 ml

Procedure:


  • Heme is gently heated under alkaline conditions in the presence of a reducing sugar such as glucose.
  • Pink needle shaped crystals of pyridine ferriprotoporphyrin or Haemochromogen are formed.
